Lacie question

I've just purchased a LaCie 250 GB D2 Extreme to use primarily as a drive to store all future virtual instrument samples. Is it better to partion this size of drive or doesn't it make a difference when used only for sample storage.

It doesn't make a difference, it's mainly for storage organisation. There are partition techniques to control which data is going to be read faster - not necessary in your case. I wouldn't even worry about it and store everything on one partition.
