For other things such as, for example, detect the tempo curve of an existing recording, or the tempo curve of something you record yourself. That, on the other hand, is something that is covered in my 10.5 book: page 231 "Recording without a metronome", or page 394 "Setting a Project Tempo by Detecting the Tempo of a Recording".

It's up to you, and depends on your preferred workflow: you can leave Flex on or you can bounce the region in place if you want to commit the Flex editing to a new audio file and turn Flex off.
